References | CHIR99021 is a cell-permeable small molecule that selectively inhibits glycogen synthase kinase 3 (GSK3)1. CHIR99021 enhances survival of mouse embryonic stem (ES) cells at low cell density, and also suppresses neural differentiation while promoting non-neural differentiation2. CHIR99021 can enable reprogramming of mouse embryonic fibroblasts transduced by Oct4 / Klf4. In addition, when combined with tranylcypromine, an inhibitor of lysine-specific demethylase 1, CHIR99021 is able to generate human-induced pluripotent stem cells in the absence of the transcription factor Sox23. The combination of CHIR99021 and SB431542 can induce uniform neural induction from human embryonic stem cells and maintain the in vitro long-term self-renewal of primitive neural stem cells 4. CHIR-99021 inhibits GSK-3 with IC50 at 7 nM. CHIR-99201 does not exhibit cross-reactivity against cyclin-dependent kinases (CDKs) with 350-fold selectivity toward GSK-3β compared to CDKs .
