多糖裂解酶（Polysaccharide Lyase）是一类主要作用于阴离子多糖,通过β消除机制降解糖苷键形成在232nm波长有吸收的C4-C5双键的不饱和寡糖。作为一类生物技术工具常用于多糖的纯度和结构分析，以及寡糖的制备。肝素酶可选择地剪切硫酸化肝素聚糖中葡萄糖胺和糖醛酸之间α(1-4)糖苷键,三种不同的肝素酶(酶Ⅰ、酶Ⅱ、酶Ⅲ)具有非常不同的专属性。

肝素酶 (heparinase)可降解肝素和硫酸乙酰肝素，肝素酶Ⅱ主要在己糖胺和糖醛酸(葡萄糖醛酸和艾杜糖醛酸)的1-4连接位点剪切硫酸肝素和肝素,相对活性大约2:1，剪切反应的主要产物是二糖。肝素(heparin)和硫酸乙酰肝素(heparan sulfate)属于糖胺聚糖(glycosaminoglycans, GAGs),为直链多糖混杂物,经由一个糖醛酸(D-葡萄糖醛酸或L-艾杜糖醛酸)和一个D-葡萄糖胺/N-乙酰葡萄糖胺组成的重复二糖单元连接而成。每个单糖残基上可发生不同程度的硫酸化,可包含零至三个硫酸基。
